Multiplex tissue image of the month - CYLC1 in testisSperm head structural protein cylicin 1 (gene: CYLC1) is visualized in late stage spermatogenesis with multiplex immunohistochemistry (mIHC/IF). The mIHC/IF spermatids panel shows spatial localization in various cell states of late stage spermatogenesis in testis, characterized by the formation of the elongated mature sperm with its distinctive head and tail structures. With the help of this panel, cylicin 1 (CYLC1) - a cytoskeletal protein associated with the 'head' structure of the sperm - is mainly localized (green) to the cytoplasm of cell state 2 (red) and 3 (white) of late spermatids, while not present in the different cell states of early spermatids (magenta and cyan). The spatial expression pattern of CYLC1 is supported by single-cell RNA sequencing data which shows group enriched expression in spermatids.
